Proliferative insults to the liver, whether derived from hepatitis, genetic abnormalities, or liver fibrosis, generate a regenerative response that can exacerbate damage or progress to cirrhosis and hepatocellular carcinoma. Current therapeutic options for liver fibrosis include restoration of the gut barrier, inhibition of transforming growth factor-β, and anti-inflammatory agents. Based on our studies of the molecular events that regulate liver regeneration, we have developed a strategy to boost liver regeneration and thereby ameliorate liver fibrosis. This strategy exploits the central role of hepatocytes in the inflammatory response. We have found that induction of a c-jun terminal phosphorylation in hepatocytes via the TLR4 and TRIF-dependent inflammatory response initiates the proliferative response of hepatocytes during liver regeneration, and that subsequent activation of NF-κB and mitogen-activated protein kinases in hepatocytes supports liver regeneration. Furthermore, we have shown that natural dietary polyphenols have the potential to boost this regenerative response, providing a potential therapeutic approach to liver fibrosis.Q:
